**Background**
Anaplastic thyroid carcinoma (ATC) is one of the most aggressive forms of thyroid cancer, characterized by rapid growth, early metastasis, and a very poor clinical prognosis. Due to its highly invasive nature and resistance to conventional therapies, identifying novel molecular targets is critical for improving patient outcomes. Peroxisome proliferator-activated receptor gamma (PPARγ) has emerged as a significant target in cancer research, as its activation can modulate cell proliferation and induce cell cycle arrest in various malignancies. In the context of Inolitazone Cancer research, targeting PPARγ provides a potential pathway to inhibit the growth of aggressive thyroid tumor cells. Therefore, we will introduce a novel high-affinity PPARγ agonist – Inolitazone.
**Definition**
Inolitazone (also known as Efatutazone dihydrochloride) is a high-affinity PPARγ agonist with an IC50 value of 0.8 nM for growth inhibition.
**In Vitro and In Vivo Studies**
According to the Inolitazone description, this compound specifically activates PPARγ without affecting PPARα or PPARδ. In terms of Inolitazone in vitro activity, the compound upregulates the cell cycle kinase inhibitor p21 WAF1/CIP1; notably, silencing p21 WAF1/CIP1 renders cells insensitive to the drug. In transient transfection assays using a PPRE response element fused to a luciferase reporter gene (PPRE3-tk-luc), a 10 nM dose of Inolitazone activates PPARγ:RXRα-dependent transcription. In DRO cells, Inolitazone demonstrated superior potency compared to other agonists, with an EC50 of 1 nM and an IC50 of 0.8 nM, whereas Rosiglitazone showed an EC50 of 65 nM and Troglitazone showed an EC50 of 631 nM.
Regarding Inolitazone in vivo efficacy, the compound has shown significant antiproliferative activity in athymic nude mice. When administered in the diet prior to DRO tumor cell implantation, Inolitazone inhibited tumor growth in a dose-responsive manner; at the highest dose (0.025%), growth was inhibited by 94.4% by day 32, with 50% of animals failing to develop demonstrable tumors. Furthermore, when treatment began one week after implantation, Inolitazone inhibited tumor growth by 68.9% in DRO tumors and 48.3% in ARO tumors by day 35. Additionally, Inolitazone combined with Paclitaxel demonstrated additive antiproliferative activity in cell culture and minimal ATC tumor growth. In conclusion, Inolitazone is a potent and selective PPARγ agonist that inhibits the growth of anaplastic thyroid carcinoma through a p21 WAF1/CIP1-dependent mechanism.
